Alabama Jack's is a must.
Snapper's is also very good.
Somebody else mentioned the Island Grill. It's outstanding. Nice little secluded place with cheap beer and great food.

Of course as already stated, Alabama Jacks Conch Fritters are a must.

Although the commercialism of Holiday Isle Resort & Marina in Islamorada will make you want to pass it by quickly, pull over and go have a drink at Rumrunners Island Bar. It's like having a drink on a pirate ship made of Tiki Huts. Very cool scenery!
